Mechanism of action

NNMT Enzyme Inhibition

5-Amino-1MQ acts as an inhibitor of nicotinamide N-methyltransferase (NNMT), an enzyme that normally uses S-adenosylmethionine (SAM) to methylate nicotinamide and produce 1-methylnicotinamide (1-MNA). By inhibiting NNMT, 5-Amino-1MQ reduces formation of 1-MNA and alters the way nicotinamide and methyl-donor resources are used within the cell. This enzyme-level activity makes 5-Amino-1MQ useful for studying NNMT-dependent metabolic pathways.

NAD⁺ Salvage and SAM Availability

NNMT inhibition can preserve nicotinamide for use in the NAD⁺ salvage pathway while reducing consumption of SAM during nicotinamide methylation. In cultured adipocytes, 5-Amino-1MQ treatment has been associated with increased intracellular NAD⁺ and SAM concentrations alongside reduced 1-MNA. These findings make the compound useful for studying the relationship between NNMT activity, NAD⁺ metabolism, and cellular methyl-donor balance.

Adipocyte Metabolism and Lipogenesis

Preclinical research has also examined how NNMT inhibition affects adipocyte metabolism. In cultured adipocytes, 5-Amino-1MQ was associated with reduced lipogenesis, while animal studies of NNMT inhibition reported changes in adipose mass and adipocyte size. 5-Amino-1MQ is therefore used in research models examining the relationship between NNMT activity, NAD⁺ and SAM metabolism, lipid storage, and broader metabolic regulation. These findings remain preclinical and do not establish corresponding metabolic effects in humans.
