Semax

Semax is a synthetic heptapeptide analog of the adrenocorticotropic hormone fragment ACTH(4-10), supplied as a high-purity lyophilized powder. Its amino acid sequence is Met-Glu-His-Phe-Pro-Gly-Pro, often abbreviated MEHFPGP. In research settings, Semax is used to study neurotrophin expression, CNS signaling, neuroprotection models, and peptide effects on BDNF, NGF, monoaminergic signaling, and stress-response pathways. Semax includes a C-terminal Pro-Gly-Pro sequence that distinguishes it from the native ACTH(4-10) fragment and is associated with improved peptide stability.

Mechanism of action

ACTH(4-10) Analog Profile

Semax is derived from the ACTH(4-10) peptide region but replaces the C-terminal portion of native ACTH(4-10) with Pro-Gly-Pro. This gives Semax the sequence Met-Glu-His-Phe-Pro-Gly-Pro. In research models, Semax is studied as a neuroactive ACTH-fragment analog rather than as a corticosteroid or direct adrenal hormone.

Neurotrophin Expression

Semax is widely studied for its effects on neurotrophin-related signaling. Preclinical research has examined changes in brain-derived neurotrophic factor, nerve growth factor, and related receptor systems after Semax exposure. This makes Semax useful in models focused on neuronal survival, synaptic plasticity, adaptive CNS signaling, and neurotrophic pathway regulation.

BDNF / TrkB Pathway Research

Research has reported Semax-associated changes in BDNF expression and TrkB-related signaling in brain-region-specific models. BDNF is a key neurotrophic factor involved in neuronal survival, plasticity, and stress adaptation. Because of this, Semax is often used in studies examining how ACTH-fragment analogs influence BDNF-linked CNS pathways.
